摘要
目的:探讨绿茶提取物表没食子儿茶素没食子酸酯(EGCG)对结肠癌细胞株LoVo细胞和SW480细胞的作用及其可能的机制。方法:采用不同浓度的EGCG(0、10、20、35μg/mL)对LoVo细胞和SW480细胞进行处理,MTT法检测细胞增殖情况;流式细胞仪检测细胞凋亡和细胞周期变化;实时荧光定量PCR检测细胞HES1与JAG1基因的表达情况。计量资料采用采用t检验,计数资料采用χ2检验。结果 10、20、35μg/mL EGCG对LoVo细胞和SW480细胞增殖均具有抑制作用,抑制率均在EGCG浓度为35μg/mL时达到最高,其抑制作用且呈浓度依赖性,不具有时间依赖性。经0μg/ml ECCG处理后LoVo细胞、SW480细胞凋亡率分别为7.72%,16.43%。经20、35μg/mL ECCG处理后,LoVo细胞和SW480细胞凋亡率分别为15.1%,19.13%和21.4%,40.81%,与0μg/mL ECCG处理后比较,差异有统计学意义((χ2=6.765,19.702,P<0.05)。35μg/mL EGCG能将LoVo细胞和SW480细胞的细胞周期阻滞在GO/G1期,阻碍其向S期转换。35μg/mLEGCG能下调LoVo细胞HES1基因和JAG1基因的表达水平(t=7.686,10.92,P<0.05)。结论:EGCG对体外培养的LoVo细胞和SW480细胞增殖有明显的抑制作用,且能诱导细胞凋亡和影响细胞周期。其作用机制可能与下调HES1及JAG1的基因表达及激活Notch信号转导途径有关。
Objective :To study the inhibitory effect of epigallocatechin- 3 -gallate (EGCG) on LoVo cells and SW480 cells and the mechanism of inhibit eolorectal cancer. Methods : Colorectal cancer cells were cultured and the proliferation ability of cul-tured cells in varying concentrations of EGCG ( 10 μg/tnL,20μg/mL,35 μg/mL) were tested by MTT analysis, and the cell apop- tosis and ceils cycle were detected with flow cytometry, and the gene expressing of HES1 and JAG1 were detected with Real - time PCR. Results :The inhibition of EGCG (tested by MTT analysis) to LoVo cells and SW480 cells had a dose -dependent manner, but didnt have a time -dependent manner. EGCG (detected with flow cytometry) can enhance the LoVo cells and SW480 cells' apoptosis rate. At the mean time, EGCG can block the cell cycle of SW480 ceils and LoVo cells at G0/G1 phase, and suppress their proliferation, EGCG can down -regulate the expression of HES1 and JAG1 gene of LoVo cells,but the changes of the expres- sion of HES1 and JAG1 of SW480 cell lines were not significant. Conclusion : EGCG can significantly inhibit the proliferation of SW480 ceils and LoVo cells by changing cell cycle and inducing cell apoptosis. The mechanism may be related to activating the gene expression of HESI and Notch ceils signaling delivery.
