摘要
目的探讨miR-127、miR-155在EB病毒(epstein-barr virus,EBV)阳性/阴性的儿童散发性伯基特淋巴瘤(Burkitt’s lymphoma,BL)中的表达。方法采用原位杂交和qRT-PCR技术检测40例儿童BL及15例扁桃体组织2株细胞株(Raji和Ramos)的EBER及miR-127、miR-155表达情况。结果 (1)40例BL儿童中16例EBER阳性,15例扁桃体EBER均阴性。(2)miR-127在EBV阳性的BL中的表达(2.68±2.95)比EBV阴性的BL(0.36±0.24)及扁桃体组织(1.23±4.09)高,差异有显著性(P<0.05);Raji和Ramos比较,miR-127表达显著升高(t=-13.038,P=0.000)。(3)miR-155在EBV阳性BL(22.50±56.84)和阴性BL(15.89±51.57)中表达差异无显著性(P>0.05);与扁桃体组织相比表达(0.31±0.54)上调(P<0.05);Raji和Ramos比较,miR-155表达显著升高(t=-3.337,P=0.008)。结论 miR-127和miR-155的高表达与儿童BL的发病机制密切相关,miR-127高表达与EBV感染相关,可能起着类似癌基因的作用。
Purpose To investigate the expression of miR-127 and miR-155 in sporadic Burkitt' s lymphoma (BL) in children with positive and negative Epstein-Barr virus (EBV) infection. Methods qRT-PCR was used to detect the expression of miR-127 and miR-155 in these cases, including 16 cases of EBV-positive BL, 24 cases of EBV-negative BL, 15 case of tonsil tissue and 2 BL cell lines ( Raji and Ramos). Results Compared EBV-positive BL (2. 68 ± 2. 95) with EBV-negative BL cases (0. 36 ± 0. 24) and tonsil tissues ( 1.23 ±4. 09) , miR-127 expression was down-regulated (P 〈0.05). The expression of miR-155 in EBV-negative ( 15.89 ± 51.57 ) and EBV-positive BL (22. 50± 56.84) had no significant differences (P 〈 0. 05 ) , miR-127 expression in Raji cell was higher than that in Ramos cell (t = - 13. 038, P = 0. 000). The expression of miR-155 expression in BL compared with tonsil tissue (0. 31 ± 0. 54) was overexpressed (P 〈 0. 05); miR-155 expression in Raji cells was higher than that in Ramos cells (t = -3. 337, P = 0. 008). Conclusion Expression of miR-127 and miR-155 is closely related to the pathogenesis of EBV-associated BL.
