人热休克蛋白对树突状细胞表面共刺激分子表达影响的研究 被引量:1

摘要 目的:研究人热休克肿瘤抗原复合物对树突状细胞(dendritic cells,DC)表面共刺激分子表达的影响。方法:分离培养人外周血来源的DC细胞,在培养过程中加入分离纯化的人热休克蛋白70(HSP70)抗原复合物及肿瘤细胞裂解物,流式细胞技术分析比较负载HSP70肿瘤抗原复合物、负载肿瘤细胞裂解物及负载前DC细胞表面分子CD80、CD83、CD86及HLA-DR表达水平的变化。结果:培养基中加入HSP70肿瘤抗原复合物后,其表面共刺激分子CD80、CD86和成熟标记分子CD83的表达水平分别为(98.9±1.5)%、(98.3±1.3)%和(53.1±0.8)%,与负载前CD80、CD86和CD83的(66.2±1.6)%、(94.2±2.0)%和(16.3±1.4)%相比显著提高,P=0.013;与肿瘤细胞裂解物负载的DC表面CD80、CD86和CD83的(86.2±1.9)%、(96.5±1.1)%和(55.1±1.0)%相比,共刺激分子CD80的表达显著提高,P=0.020。结论:人热休克肿瘤抗原复合物可提高DC表面共刺激分子的表达水平,促进DC功能活化,有可能在抗肿瘤免疫机制中起重要作用。 OBJECTIVE:To study the effect of human shock protein on dendritic cell(DCs)surface costimulating molecules expression.METHODS:DC cells were isolated and cultured from peripheral blood,loaded by HSP70peptides complex and tumor cells lysates,and then the expression of DC cells membrane surface molecular CD80,CD83,CD86and HLA-DR were detected by flow cytometry before and after loaded.RESULTS:The expression of co-stimulatory molecular CD80,CD86and mature labeled molecular CD83of DC cells loaded with HSP70peptides complex were(98.9±1.5)%,(98.3±1.3)%and(53.1±0.8)%,which were raised by controlled with DC before loaded(66.2±1.6)%,(94.2±2.0)%and(16.3±1.4)%(P=0.013),and controlled with DC loaded with tumor cells lysates(86.2±1.9)%,(96.5±1.1)%and(55.1±1.0)%.CD80was raised(P=0.020).CONCLUSION:It is suggested that Hsp70peptide complex can improve the expression of co-stimulatory molecular of DCs,activate DC functions directly,which may possess important effect in immunologic anti-tumor mechanism.
