结缔组织生长因子在颅内动脉瘤壁上的表达

摘要 目的通过研究结缔组织生长因子(CTGF)在颅内动脉瘤壁上的表达来探讨CTGF对颅内动脉瘤形成发展的影响。方法中国医科大学附属第一医院神经外科2007年12月至2011年12月收治动脉瘤中的16例动脉瘤病例,以颞浅动脉作对照组,通过实时逆转录酶-聚合酶链式反应(RTPCR)检测及免疫组化进行研究,并予统计学分析。结果定量检测RT-PCR表明对照组(STA)比较试验组(IA)CTGF表达明显升高,差异有统计学意义(P<0.05)。免疫组化显示动脉瘤壁中膜和外膜CTGF表达阳性率较内膜高(χ2=9.94,P<0.05)。结论 CTGF作为一个特异性的靶位,将会有可能对预防和治疗颅内动脉瘤提供新的思路。 Objective To study the effect of CTGF on the formation of intracranial aneurysms (IAs) by analyzing the expression of connective tissue growth factor (CTGF) on the wall of intracranial aneurysm. Methods Sixteen tissue samples of intracranial aneurysms were taken randomly from December 2007 to December 2011 in Department of Neurosurgery, First Affiliated Hospital of China Medical University. Superficial temporal artery (STA) was served as the control group. The pathological changes and the expression of CTGF were analyzed by Western blot and real time reverse transcription-polymerase chain reaction (RT-PCR), respectively. Results CTGF expression of the control group (IA/STA) was significantly increased compared with that of the experimental group (IA) by the Western blot and semi-quantitative RT-PCR, and the difference was statistically significant (P 〈 0. 05 ). Immunohistochemistry showed that CTGF-positive rate of the adventitia and tunica media of ameurysm wall was higher than that of endometrium (X2 =9.94, P 〈0.05). Conclusion CTGF, as a specific target of intracranial aneurysms, will has the potential to provide new ideas for prevention and treatment of intracranial aneurysms.
