香菇多糖联合多西他赛对胃癌BGC823细胞增殖的影响 被引量:8

摘要 目的:观察香菇多糖单药及联合多西他赛抑制胃癌细胞BGC823增殖的作用。方法:分别用不同浓度的香菇多糖、多西他赛和香菇多糖联合多西他赛处理胃癌细胞BGC823。香菇多糖终浓度分别为1.560μg/ml,3.125μg/ml,6.250μg/ml,12.500μg/ml。多西他赛终浓度分别为0.625μg/ml,1.250μg/ml,2.500μg/ml,5.000μg/ml。MTT法检测各组细胞增殖情况;同时用Annexin V/PI法检测各组细胞的凋亡。结果:单药香菇多糖能抑制BCG823细胞的增殖,不同浓度(由高到低)对BGC823细胞的增殖抑制率分别为67.7%,56.0%、42.1%和23.2%,与阴性对照组比较有统计学意义(P<0.05)。不同浓度香菇多糖联合多西他赛能增强多西他赛对BGC823细胞增殖的抑制作用。单药香菇多糖能增加BGC823细胞的细胞凋亡率,与多西他赛联用使细胞凋亡率明显增加(P<0.05)。结论:香菇多糖能抑制胃癌BGC823细胞的增殖,并能显著增强多西他赛抑制胃癌细胞BGC823的增殖作用。 Objective:To investigate whether lentinan, lentinan combined with docetaxel can inhibit the cell prolif- eration in human gastric cancer cell line BGC823. Methods:BGC823 cells were divide into lentinan, doeetaxel and lentinan combined docetaxel groups. Lentinan was added in the cuhuremedium to concentration of 1. 560μg/ml, 3. 125μg/m1,6. 250μg/ml, 12.5001xg/ml. Docetaxel was added in the culturemedium to concentration of 0. 625μg/ ml, 1. 250μg/rn1,2. 500μg/m1,5. 0001xg,/ml. MTT was applied to detect the proliferation of BGC823 cells. Apoptosis was detected by Annexin V/PI. Results: Lentinan could inhibit the proliferation of BGC823. Compared with the control group, the inhibitary rates were 67.7% ,56.0% ,42.1% and 23.2% with different concentrations of lentin- an(P 〈0.05) . The inhibitory rates of the proliferation of BGC823 ceils were significantly higher in combined with docetaxel than those in docetaxel alone group(P 〈 0.05 ). Lentinan can increase the apoptosis rates of BGC823 cells, the apoptosis rate was higher in combined with docetaxel group ( P 〈 0.05 ). Conclusion: Lentinan can inhibit the pro- liferation of gastric cancer BGC823 cells, and can significantly enhance the inhibit rate of proliferation in docetaxel.
