β-catenin异常表达在结肠癌发生发展中的作用研究 被引量:4

摘要 目的:探讨β-catenin在原发性结肠癌中表达及临床意义。方法:应用免疫组化Envision法检测β-catenin蛋白在138例结肠癌(48例含癌旁组织)、37例结肠腺瘤组织及66例结肠正常黏膜中的表达;应用实时荧光定量PCR检测β-catenin基因在37例新鲜原发性结肠肿瘤标本及其配对37例正常结肠中表达;分析β-catenin蛋白及其基因表达与临床病理的关系。结果:β-catenin在正常结肠、结肠腺瘤、癌旁组织及结肠癌胞质中阳性率分别为42.4%,64.9%,72.9%和68.8%;β-catenin蛋白表达与淋巴结转移、Duke分期有关(P<0.05)。β-catenin mRNA在结肠肿瘤中平均表达是正常结肠中的1.946倍(P<0.05),表达水平与肿瘤部位及淋巴结转移有关。结论:β-catenin因子的异常表达与结肠癌的发生发展密切相关。 Objective:Investigate the expression and clinical significance of β-catenin in primary colon cancer. Methods: The ex- pressions of β-catenin protein in 138 colon cancer tissues(48 cases with adjacent tissues of cancer) ,37 colon adenoma tissues and 66 normal colon tissues were detected by Envision immunohistochemistry. The expressions of ^-catenin mRNA in 37 colon tumor tissues and 37 matched normal colon tissues were detected by RT-PCR. The clinical and pathological features of β-catenin protein and mRNA were analyzed. Results: The respective positive rate of β-catenin protein in the cytoplasm of normal colon mucosa,colon adenoma tis- sues, adjacent tissues of cancer and colon carcinoma tissues were 42.4% ,64.9% ,72.9% and 68.8%. The expressions of β-catenin were correlated with lymph node metastases and the Duke stage ( P 〈 0.05 ). The average expression of ^-catenin mRNA in colon canc- er was 1. 946 times of normal ones (P 〈 0.05) and the expression level was correlated with lymph node metastases and tumor loca- tions. Conclusion: The abnormal expressions of 13-catenin protein are closely correlated with the development of colonic neoplasm.
