摘要
采用茎尖分生组织培养技术 ,获得了大蒜 (AlliumsativumL .)的无病毒试管苗。通过基本培养基和激素配比试验 ,筛选出最佳的培养基组成 ,进行脱病毒苗的快速繁殖。结果表明 :诱导愈伤组织的最适培养基为 :MS +BA0 .2mg/L +NAA 0 .5mg/L ,月生长率达 12 70倍 ;诱导丛生芽的最适培养基为 :B5+BA 0 .5mg/L +IAA 0 .2mg/L ,丛生芽繁殖系数高达 2 5 5倍 ,技术上达到了快速繁殖规模生产的要求。用电子显微镜反复进行了病毒检测 ,其中有 5个样品脱病毒彻底 ,将作为今后提供无病毒优质种苗的原种苗。
Virus\|free materials of \%Allium sativum \%L. were obtained from \%in vitro \%shoot apex tissue culture. A series of optimization experiment for cultural medium composition, concentration of phytohormones were investigated with a view to accelerate the propagation of virus\|free materials. The obtained results indicated that the optimized medium for inducing callus was MS+BA 0.2 mg/L+NAA 0.5 mg/L. The growth rate of callus in one month was 12.70. The best medium for multiplying clumpy buds was B\-\{5\}+BA 0.5 mg/L+IAA 0.2 mg/L, the highest propagating ratio reached 25.5. These results could meet the technology requirements of rapid\|propagation in large scale production. Virus \|free materials were observed repeatedly under electron microscope, 5 virus\|free samples were tested out which could be used to fast propagation for production.
