姜黄素通过内质网应激途径诱导宫颈癌HeLa细胞凋亡的实验研究 被引量:7

摘要 目的:观察姜黄素诱导宫颈癌HeLa细胞的凋亡并探讨其相关机制。方法:在以不同浓度姜黄素处理HeLa细胞后,采用MTT法观察细胞活性并选择姜黄素的最适干预浓度。将等量HeLa细胞分为4组:对照组(Ctrl)不加任何药物干预;对照组+4-苯基丁酸钠组(Ctrl+PBA)采用4-PBA(3mmol/L)对HeLa细胞进行干预;姜黄素组(Cur)采用姜黄素(25μM)对HeLa细胞进行干预;姜黄素+4-苯基丁酸钠组(Cur+PBA)则在姜黄素干预后48h采用4-PBA(3mmol/L)对HeLa细胞进行干预。流式细胞术检测各组HeLa细胞凋亡情况;采用Real-time PCR和Western Blotting检测各组HeLa细胞中内质网应激相关蛋白葡萄糖调节蛋白78(GRP78)以及C/EBP同源蛋白质(CHOP)mRNA及蛋白表达情况。结果:不同浓度姜黄素干预HeLa细胞后,细胞生长均受到不同程度的抑制,并呈现出浓度依赖性;与Ctrl及Ctrl+PBA组相比,Cur组HeLa细胞凋亡率显著升高;与Cur组相比,Cur+PBA组HeLa细胞凋亡率显著降低;与Ctrl组及Ctrl+PBA组相比,Cur组GRP78及CHOP mRNA及蛋白表达水平均显著升高(P<0.05);与Cur组相比,Cur+PBA组GRP78及CHOP mRNA及蛋白表达水平均显著降低(P<0.05)。结论:内质网应激途径是姜黄素诱导HeLa细胞凋亡的重要机制之一。
