MACC1基因对人小细胞肺癌细胞株SBC-5细胞生物学特性的影响

摘要 目的:采用RNA干扰(RNAi)技术探讨MACC1基因对人小细胞肺癌细胞株SBC-5细胞生物学特性的影响。方法:将siRNA-MACC1转染SBC-5细胞;利用Western blot法检测转染后MACC1蛋白的表达;利用MTT法测定细胞增殖;利用Hoechst法检测转染细胞的凋亡。结果:SBC-5细胞,转染基因特异性的siRNA后MACC1蛋白表达明显降低,凋亡明显增加,细胞增殖活力明显降低。结论:MACC1基因特异性的siRNA可抑制蛋白表达,促进SBC-5细胞凋亡并抑制细胞增殖。
