大鼠肝脏在四氯化碳作用下NF-κB与CXCR1表达的研究 被引量:3

摘要 目的:采用四氯化碳造成大鼠肝损伤,观察大鼠肝脏组织病理学变化,同时检测NF-κB和CXCR1在肝脏组织中的表达。方法:30只健康雄性SD大鼠随机分为对照组,实验组。实验组每周使用四氯化碳处理1次,分别在第3周、6周、9周取大鼠肝脏,进行HE染色,观察肝脏变化。同时分别采用实时定量PCR以及Western blotting检测肝脏组织,NF-κB和CXCR1的mRNA和蛋白表达。结果:与对照组相比,实验组大鼠肝脏随着造模时间的延长,大鼠肝纤维化程度加深,出现细胞水肿,炎症细胞增多,轻度的脂肪变化。实验组各组大鼠肝脏组织中的NF-κB和CXCR1mRNA以及蛋白表达逐渐增高。结论:大鼠经四氯化碳作用后,肝脏中的NF-κB和CXCR1表达量都有升高,机体可能是通过这种方式应对肝脏的损伤,起着修复保护作用,如果持续损伤则导致肝脏纤维化。
