摘要
目的斑马鱼具有很强的脊髓损伤后修复的能力,但是目前机制尚不明确。为研究斑马鱼脊髓修复过程中大脑对脊髓的可能作用与影响,本文采用成年斑马鱼脊髓损伤模型,检测了其脑组织中血管内皮生长因子(vascular endothelial frowth factor,vegf)基因的表达情况。方法建立成年斑马鱼脊髓损伤模型,应用实时定量PCR方法以及原位杂交技术,在成年斑马鱼脊髓损伤后4 h、12 h、6 d、11 d检测脑组织中血管内皮生长因子(vascular endothelial frowth factor,vegf)基因的表达情况。结果成年斑马鱼脊髓损伤以后,血管内皮生长因子基因损伤急性期(4 h、12 h)在脑内呈现显著性升高(P<0.05),而在神经修复期(6 d)脑内的血管内皮生长因子(vegf)基因则呈现恢复趋势,在损伤后11d此基因出现显著性下降的趋势(P<0.05)。结论成年斑马鱼脊髓损伤后,短期内高表达vegf基因可能为修复中血管的再生提供有利的刺激与内环境,随着修复时间的延长和修复的逐渐完成,此基因则呈现恢复直至下降的趋势。
Objective Plenty of studies suggested that zebrafish has a strong ability of spinal cord regeneration after severe injury, but the mechanism is obscure. In order to investigate the function of brain during the spinal cord regeneration,we detected the change of vegf gene in the brain of our zebrafish spinal cord injury models. Method Built spinal cord injury models in adult zebrafish, collected the brain tissues in 4h, 12h, 6d and l ld time course. Real -time quantitative PCR and in situ hybridization were used to detect the change of veff expression in the brain of injured zebrafish. Results Contrasted with sham group, the mRNA levels of vegf in zebrafish brain were up - reg- ulated in the acute phase (4 hours and 12 hours post - SCI) significantly (P 〈0.05 ). However, compared with the sham group, the mRNA levels of vegf in zebrafish brain were down - regulated in the chronic phase of neuroregeneration (6 days and 11 days post- SCI), especially in the 11 days, the expression level of vegf reduced significantly (P 〈 0.05). Conclusion These results suggested that brain may play a role in the process of adult zebrafish spinal cord regeneration through a modulation of vegf expression.
