全蝎纯化液对凝血酶诱导血管内皮细胞释放NO、TFPI及表达TF的影响 被引量:19

Effects of Scorpion Purified Liquid on the Release of NO,TFPI,TF Induced by Thrombin in Cultured Vascular Endothelial Cells

摘要 目的探讨全蝎纯化液对凝血酶诱导血管内皮细胞释放一氧化氮(NO)、组织因子途径抑制物(TFPI)及表达组织因子(TF)的影响。方法对传代培养的人脐静脉细胞(HUVEC)(2代~3代)分别给予不同的处理,取上清液测定NO、TFPI,取细胞冻融液测TF活性。结果与空白对照组比较,凝血酶组能明显抑制血管内皮细胞释放TFPI(P<0.01),与凝血酶组比较,全蝎纯化液各剂量组能拮抗TFPI的降低(P<0.05)。凝血酶促进血管内皮细胞表达TF,较对照组明显增高(P<0.01),全蝎纯化各剂量组较凝血酶组则明显降低(P<0.01)。全蝎纯化液各剂量组促进血管内皮细胞释放NO,较对照组、凝血酶组明显增高(P<0.01)。结论全蝎纯化液能抑制凝血酶诱导血管内皮细胞表达TF,可拮抗凝血酶抑制TFPI释放,促进血管内皮细胞释放NO,提示全蝎纯化液对凝血酶诱导血管内皮细胞的损伤具有保护作用。 Objective To observe the effect of scorpion purified liquid(SPL) on the release of nitric oxide (NO), tissue factor pathway inhibitor (TFPI), tissue factor (TF) induced by thrombin in cultured vascular endothelial cells (VEC). Methods The hu man umbilical vein endothelial celIs (HUVECs) were cultured in vitro. The well grown 2 to 3 generation HUVECs were given different treatment respectively. The supernatant fluid were taken to measure NO, TFPI,The freezing and thawing liquid were taken to measure the activity of TF. Results Compared with the blank control group, thrombin group could obviously inhibit the release of TFPI on HUVEC(P〈0.01). Compared with thrombin group,SPL in each dose group could antagonize the decrease of TFPI(P% 0.05). Thrombin promoteed the express of TF on VEC, it was significantly lower in the control group(P〈0.01). Compared with the thrombin group,SPL in each dose group was lower(P〈0.01). Compared with the blank control group and thrombin group, the con centration of NO from scorpion purified liquid each dose group was remarkably increased(P〈0.01). Conclusion SPL could inhibit the release of TF, can antagonize the inhibition of the release of TFPI from HUVEC induced by thrombin and promote the release of NO from HUVEC induced by thrombin, suggesting that scorpion protect the iniured VEC induced hv thrnrnhin
