不同H-FABP基因型滇南小耳猪脂滴蛋白相关基因表达差异研究 被引量:2

摘要 探究不同心型脂肪酸结合蛋白基因型滇南小耳猪脂滴蛋白相关基因表达量与肌内脂肪细胞甘油三酯含量的关系,为云南地方猪种肌内脂肪沉积机理提供理论依据。选取不同H-FABP基因型滇南小耳猪商品猪各8头,分离背最长肌肌内脂肪细胞,采用实时荧光定量PCR的方法,检测滇南小耳猪脂肪特异性蛋白27、肥胖基因、围脂滴蛋白1和围脂滴蛋白2基因mRNA表达量,并分析其与肌内脂肪细胞甘油三酯含量的关系。结果:H-FABP基因的HH基因型FSP27、FTO、PLIN1和PLIN2基因表达量和肌内脂肪细胞甘油三酯含量均为高于Hh型和hh型,其中HH型显著高于hh型(P<0.05);相关性分析显示FSP27和PLIN1基因mRNA表达量与肌内脂肪细胞甘油三酯含量呈强度正相关,FTO和PLIN2基因mRNA表达量与肌内脂肪细胞甘油三酯含量呈中度正相关。试验表明:FTO基因、FSP27基因、PLIN1基因和PLIN2基因的表达量不同可能是不同H-FABP基因型滇南小耳猪肌内脂肪细胞甘油三酯含量存在差异的原因之一。 The present study was to investigate the relationship between the expression level of lipid droplet genes and the triglyceride con- tent in intramuscular adipocytes in Diannan small-ear pigs with different H-FABP genotypes, which provide the theory for the mechanism of intramuscular fat deposition in Yunnan local pig breeds. Eight Diannan small-ear pigs per H-FABP genotype were selected. Intramuscular adipocytes were isolated from longissimus dorsi muscle. Real-time PCR was performed to quantify the expression levels of FSP'27, FTO, PLIN1 and PLIN2 genes, then analyzed the correlation coefficients between the triglyceride content and the expression level of mRNA in intramuscular adipocytes. The results showed that the expression level of lipid droplet genes, including FSP27, FTO, PLIN1 and PLIN2 was significantly increased in Hinf I PCR-RFLP HH compared with other genotypes ( P〈0.05 ). The triglyceride content was significantly increased in Hinf I PCR-RFLP HH compared with other genotypes (P〈0. 05). Correlation analysis showed that the expression level of FStE7 and PLIN1 genes were high positively correlated with the triglyceride content. The expression levels of FTO and PLIN2 were moderate positively correlated with triglyceride contents. Together, the difference of triglyceride contents in intramuscular adipocytes might come from the expression levels of FFO, FSP27, PLIN1 and PLIN2 genes.
