摘要 采用超临界CO2诱变淀粉酶产生菌枯草芽孢杆菌(Bacillus subtilis),提高其产淀粉酶的活力。结合平板初筛与发酵培养测定酶活力复筛的方法,考察了诱变条件如诱变压力、诱变温度和诱变时间对诱变该菌的影响。其最佳诱变条件为:压力8.0 MPa、温度37℃、时间30 min,在此条件下,诱变株的淀粉酶活力提高了约33%。 Mutation breeding of amylase-producing strain Bacillus subtilis K2 using supercritical CO2 was investigated.Based on the plate method and determination of amylase activity,the mutation conditions including pressure,temperature and time were examined.The optimal mutation conditions were as follows: pressure 8.0 MPa,temperature 37℃,time 30 min.A mutant with almost 33% increase in amylase activity compared with the wild type strain was isolated.
