反相高效液相色谱法测定酸枣叶中芦丁的含量

Determination of Rutin in Leaves of Ziziphus jujuba var. spinosa by HPLC

建立了高效液相色谱法测定酸枣叶中芦丁含量的方法。采用Kromasil C18色谱柱;以甲醇-0.1%磷酸(50:50)为流动相;流速为1 mL/min;检测波长为370 nm测定了忻州地区酸枣叶中芦丁含量。结果表明:芦丁回归方程y=675 221x+794.16(r=0.999 8),在0.153μg～1.53μg范围内线性关系良好,加样回收率100.57%,RSD=0.96%(n=5)。该法操作简便、准确,重复性好,可用于酸枣叶中芦丁含量测定。

The contents of rutin in leaves of Ziziphus jujuba var. spinosa were determined by high performance liquid chromatography （HPLC）. The samples was separated using Kromasil C18 （150 mmx4.6 ram, 5 μm） column, Methanol -0.1%phosphoric acid （50 ： 50） as mobile phase at a flow rate of 1.0 mL/min and detection wavelength of 370 nm. The result suggested that the linear range of rutin concentration was from 0.153μg to 1.53 μg. y=675 221x＋794.16 （r=0.999 8）, the average recovery was 100.57 % with the RSD of 0.96 %. The proposed method had highly repeatability and selectivity. It was simple, fast and could be used for the determination of rutin in leaves of Ziziphus jujuba var. spinosa.
