摘要
目的根据采供血机构实际工作需要,选择滤除白细胞的可靠时间点,确保滤白血制品在保存期内安全、有效地用于临床。方法通过对(4±2)℃、6h内保存和(4±2)℃、24h内保存的全血各30份,进行占线法过滤后,测定白细胞滤除率、红细胞回收率,以及保存末期的游离血红蛋白含量。结果经占线法过滤后,6 h内过滤的白细胞清除率达99.95%,红细胞回收率达92.98%;24 h内过滤的白细胞清除率达99.98%,红细胞回收率达90.69%;两者过滤后白细胞残留量的差异具有统计学意义(t=9.52,P<0.01),红细胞回收率的差异无统计学意义(t=1.53,P>0.05);滤白后保存末期红细胞溶血率为0.10%,与对照组比较差异无统计学意义(t=1.89,P>0.05)。结论全血在滤除白细胞时的冷藏时间明显影响白细胞的滤除效果,但对红细胞回收无明显影响。
Objective According to the requirement of blood service institution for practical work, to select the de- pendable time for white blood cell filtration and ensure the safety and effectiveness of leukocyte-free products for clinical application during preservation period. Methods To determine the white blood cell clearance rate, the red blood cell recov- ery rate and the content of free hemoglobin at the end of preservation, by filtrating each of 30 whole blood samples pre- served for 6 h and 24 h at (4±2)℃ respectively. Results After in-line filtration, the white blood cell clearance rate reached 99. 95% and the red blood cell recovery rate was 92.98% within 6 h; the white clearance rate made 99.98G and the red recovery rate was 90.69% within 24 h. The difference between the two white blood cell residues was statistical sig- nificant (t = 9.52,P〈0. 01), but it wasn't that between the two red blood cell recovery rates (t = 1.54,P〉 0. 05). The hemolysis rate of red cell was 0.10% at the end of preservation with white cell filtrated, which was no statistical significant compared with the control group (t= 1.89,P〉0.05). Conclusion The refrigeration time of whole blood in white blood cell filtration has apparent impact on white blood cell clearance, but little on red blood cell recovery.
