人脐带间充质干细胞对NOD/SCID小鼠乳腺原位移植人乳腺癌MCF-7细胞增殖分化的影响 被引量:3

摘要 目的在NOD/SCID小鼠乳腺原位移植人乳腺癌MCF-7细胞的模型中,研究人脐带间充质干细胞(MSCs)对人乳腺癌MCF-7细胞所形成的异种移植物的增殖和分化情况。方法分别在NOD/SCID小鼠乳腺原位移植人乳腺癌MCF-7细胞、第1代人脐带MSCs或两者混合物,然后处死小鼠,剥离所形成的异种移植物,做病理切片并观察异种移植物的组织形态学特征。结果人乳腺癌MCF-7细胞的异种移植物成瘤率低,人脐带MSCs促进异种移植物的形成并分化成具有人乳腺结构样组织学特征。结论人脐带MSCs具有促进人乳腺癌MCF-7细胞在NOD/SCID小鼠乳腺原位形成具有人乳腺样结构的作用。 Objective To implant breast cancer cells in an orthotopic site along with human mesenchymal stem cells (MSCs) in NOD/SCID mice, in order to study the growth and differentiation of human breast cancer MCF-7 cells. Methods MCF-7 cells, MSCs or blend of them were injected into mouse mammary fat pad of the fourth nip- pie. The mice were executed, and the tumors were collected and examined for morphology. Results MCF-7 cells were weakly tumorigenic, human MSCs could promote the formation of xenografts and differentiating into the human mammary gland-like structures. Conclusion Human umbilical cord MSCs can promote human breast cancer MCF- 7 cells differentiating into the human mammary gland-like structures.
