摘要
目的观察白藜芦醇对60Coγ射线照射小鼠造血系统损伤修复的影响。方法 ICR小鼠随机分成5组,即正常对照组、照射对照组以及白藜芦醇50、100、200 mg.kg-1照射组,照射前3 d灌服给药,每天1次。除正常对照组外,所有小鼠60Co射线全身5.5 Gy辐照1次,照射后第7天小鼠取骨髓细胞和脾脏,对骨髓单个核细胞和脾集落形成单位进行计数,测定骨髓细胞DNA含量和细胞周期。结果与正常对照组比较,照射对照组小鼠骨髓单个核细胞数和骨髓细胞DNA含量显著降低(P<0.05),脾集落形成单位数和S和G2/M期细胞百分率显著增多(P<0.05);与照射对照组比较,白藜芦醇100、200 mg.kg-1照射组小鼠照射骨髓单个核细胞数、骨髓细胞DNA含量、脾集落形成单位数和S和G2/M期细胞百分率显著增多(P<0.05),白藜芦醇50 mg.kg-1照射组有增多趋势。结论白藜芦醇可以促进60Coγ射线照射小鼠造血系统损伤的恢复,防护效果与给药剂量相关。
Objective To investigate the radioprotective effects of resveratrol(RES) on the recovery of hematopoiesis in irradiation-injured mice.Methods Thirty male ICR mice were randomized into 5 groups,including normal control group,irradiated control group,RES 50 mg · kg-1 irradiated group,RES 100 mg · kg-1 irradiated group and RES 200 mg · kg-1 irradiated group.RES was administered respectively to three dosage groups by gavage for 3 days before irradiation.Except for normal control,all mice were whole-body irradiated by 60Co γ ray(a single dose of 5.5 Gy).The mice were killed on 7th day after irradiation.The number of bone marrow mononuclear cells(BMNC) and colony forming units-spleen(CFU-S) was counted.The content of DNA of marrow cells and and cell cycles of bone marrow cell were measured.Results Compared with normal control group,the number of bone marrow mononuclear cells(BMNC) and the content of DNA of marrow cells were significantly decreased(P0.05) and the phase S and G2/M marrow cell percentage and the number of CFU-S were significantly increased in irradiated control group mice(P0.05).Compared with irradiated control,the number of bone marrow mononuclear cells(BMNC),the content of DNA of marrow cells,the phase S and G2/M marrow cell percentage and the number of CFU-S were significantly increased in 100 and 200 mg · kg-1 RES irradiated groups(P0.05).There were similar changes in 50 mg · kg-1 RES irradiated groups.Conclusion RES can promote the recovery of hematopoietic system injury in 60Coγ rays irradiated mice and the radioprotective effects of RES are correlated with the dosage.
