摘要
目的:观察独活寄生汤治疗前后膝骨关节炎患者Wnt5a、β-catenin、BMP-2 mRNA表达的变化,探讨独活寄生汤对膝骨关节炎的作用机制。方法:42例膝骨关节炎患者,予独活寄生汤治疗12周,应用VAS法记录患者治疗前后主要症状变化,应用实时荧光定量PCR技术检测治疗前后Wnt/β-catenin-BMP信号通路中相关指标变化。结果:治疗后KOA患者VAS积分明显降低(P<0.05);Wnt-5a、β-catenin、BMP-2 mRNA表达较治疗前呈下降趋势,与治疗前比较,有统计学意义(P<0.05)。结论:独活寄生汤可能通过介导Wnt/β-catenin信号通路,抑制其下游靶基因BMP-2转录,保护关节软骨,发挥缓解疼痛的治疗效用。
Objective: To explore the mechanism of duhuo jishen decoction on knee osteoarthritis by observing the changes of Wnt5a,β- catenin and BMP- 2mRNA before and after treatment.Methods: 42 Cases with knee osteoarthritis were treated with duhuo jishen decoction for 12 weeks,observing the total curative effect and VAS score,changes of Wnt5a,B- catenin and BMP- 2mRNA were determined by real- time PCR.Results: The total effective rate was 80. 95%,the VAS score in knee osteoarthritis patients was decreased obviously after treated with duhuo jishen decoction,the expression of Wnt5a,B- catenin and BMP- 2mRNA was lowered significantly after treated.Conclusion: Duhuo jisheng decoction can effectively treat knee osteoarthritis by inhibiting BMP- 2 expression through Wnt /β- catenin signal pathway.
