升麻鳖甲汤对MRL/lpr小鼠Th_1/Th_2细胞因子失衡的影响 被引量:5

EFFECTS OF SHENGMA BIEJIA DECOCTION ON IMBALANCE OF Th_1/ Th_2 IN MRL / lpr MICE

摘要 目的:观察升麻鳖甲汤对MRL/lpr小鼠Th1/Th2细胞因子失衡的影响。方法:选用24只雌性MRL/lpr小鼠随机分为中药组、西药组、中西药结合组、模型对照组,另取6只雌性BALB/C小鼠为正常对照组;分别予升麻鳖甲汤、强的松、强的松合升麻鳖甲汤、生理盐水灌胃,给药6周。6周后取血检测干扰素(IFN-γ)、白细胞介素(IL-12、IL-10、IL-4)水平,并观察小鼠肾脏病理变化。结果:各治疗组小鼠血清IFN-γ、IL-12水平无显著变化(P>0.05);与模型组比较,各药物组血清IL-10、IL-4水平显著降低,血清IFN-γ/IL-4比值升高,差异有统计学意义(P<0.05或P<0.05),中西药组能显著升高血清IL-12/IL-10比值(P<0.05);各治疗组均能不同程度改善肾组织病理形态及超微结构变化。结论:升麻鳖甲汤对系统性红斑狼疮的治疗作用可能与其能调节Th1/Th2细胞因子之间的失衡、改善肾组织炎性病理改变有关。 Objective: To observe the effects of shengma biejia decoction on imbalance of Th1/ Th2in MRL / Ipr mice.Methods: Twenty- four female MRL /lpr mice were randomly divided into four groups,Chinese medicine group( shengma biejia decoction),western medicine group( prednisone),Chinese and western medicine group( shengma biejia decoction + prednisone) and model group( saline),six female healthy BALB / C mice were set as normal control group( saline),each group was given the corresponding medicine for 6 weeks. Serum interferon gamma( IFN- γ),interleukin 12( IL- 12),interleukin 10( IL- 10) and interleukin 4( IL- 4) levels were detected by ELISA,the renal pathological changes in mice were also observed by light microscope.Results: There were no significant changes on serum IFN- γ and IL- 12 levels in the three medicine groups,to compare with model group,the serum IL- 10 and IL- 4 levels were decreased obviously in the three medicine groups,serum IL- 12 / IL- 10 ratio was raised,the difference between model and medicine groups was significant,the renal pathological changes were relieved in different degrees in the three medicine groups.Conclusion: Shengma biejia decoction can effectively treat systemic lupus erythematosus,its mechanism may be related to regulate the imbalance of Th1/ Th2and improve the renal pathological changes.
