摘要
目的:研究西施舌多糖对人食管癌细胞株EC-9706的增殖及抗氧化活性的影响。方法:体外培养人食管癌细胞株EC-9706,用MTT法研究不同浓度西施舌多糖对人食管癌细胞增殖的影响,并检测不同浓度西施舌多糖对体外培养食管癌细胞和小鼠原代培养肝细胞的超氧化物歧化酶(SOD)和过氧化氢酶(CAT)活性,以及对细胞培养上清液总抗氧化活性(T-AOC)和抑制羟自由基(·OH)活性的影响。结果:西施舌多糖对食管癌细胞株EC-9706增殖的抑制作用随浓度增大和培养时间延长而增加。西施舌多糖能提高体外培养细胞的抗氧化活性,而且在西施舌多糖浓度相同的条件下,小鼠肝细胞SOD、CAT、T-AOC和抑制羟自由基活性均高于食管癌细胞。结论:西施舌多糖能抑制人食管癌EC-9706细胞的增殖,增强体外培养细胞的抗氧化活性。
Objective, Anti-proliferation and antioxidant activities of polysaccharides from Coelomactra antiquata (PCA)on human esophageal carcinoma EC-9706 cells in vitro were observed. Methods: EC-9706 cells were cultured with different concentration of PCA in vitro.The proliferation inhibiting of esophageal carcinoma cells were determined by MF[ assay.Furthermore,effect of different concentration of PCA on superoxide dismutase(SQD), catalase(CAT) of esophageal carcinoma cells and mouse hepatocyte were measured, meanwhile,total antioxidant activity(T-AOC) and scavenging effect of hydroxy radical (. OH)of culture supematan were evaluated. Result= EC-9706 cell proliferation rate gradually increased with the polysaccharides concentration and time increased.PCA could enhance the activities of cells cultured in vitro.Under the condition of same polysaccharide concentration,the activities of SOD, CAT, T-AOC and scavenging ~ OH of mouse hepatocyte were all higher than esophageal carcinoma cells. Conclusion:PCA could restrain proliferation of human esophageal cancer cells,and enhance the activities of ceils cultured in vitro.
