羚羊角粉对创伤性脑水肿影响的实验研究 被引量:5

摘要 目的研究羚羊角粉对大鼠脑创伤后脑水肿的影响。方法 SD大鼠100只,随机分成模型组与治疗组,每组再按创伤前1 d,创伤后1、3、5、7 d分为5个亚组。各组大鼠复制自由落体大鼠脑外伤模型,治疗组大鼠造模后给予羚羊角粉300 mg/(kg·d)灌胃。各组大鼠分别在创伤前1 d,创伤后1、3、5、7 d处死,分别测量脑含水量;血脑屏障的功能,观察各组脑组织的病理结构以及水通道蛋白-4(AQP4)的表达。结果治疗组较模型组相比脑水肿明显减轻(P<0.05),血脑屏障损伤较少(P<0.05)。创伤后大鼠脑组织AQP4的表达升高(P<0.05),第3日达最高水平(P<0.05),治疗组AQP4的表达较创伤组有显著下调(P<0.05)。结论羚羊角粉对大鼠创伤性脑水肿有治疗作用,可能是通过抑制创伤后脑组织周围的AQP4的表达来实现的。 Objective:To study the influence of antelope horn powde on brain edema after traumatic brain injury in rats.Methods:100 SD rats were randomly divided into 2 groups including the trauma group and the antelope horn powder feeding group.Each group was divided into 5 time points:before trauma 1d and after trauma 1,3,5,7 d,each point with 10 rats.The traumatic brain injury models in rats were made through free fall.Among them,antelope horn powder feeding group rats were given antelope horn powder with 300 mg/(kg·d) by gavage.The rats were in trauma before 1 d,after trauma 1,3,5,7 d and were measured respectively such as the water content of brain,the blood brain barrier function,the cerebral tissue pathological structure and the AQP4 expression.Results:Compared with trauma group,the brain water was significantly reduced (P< 0.05),and blood brain barrier were less injury obviously (P<0.05) in the antelope horn powder feeding group.The increased expression of AQP4 in brain tissue of rats after traumareached the highest level in the 3rd day (P< 0.05).The AQP4 expression of the antelope horn powder feeding group was significantly lower than it of the trauma group (P< 0.05).Conclusion:The antelope horn powder has therapeutic effect on traumatic brain edema in rats.It may be through inhibition of expression of the AQP4 around traumatic brain tissue to achieve these effect.
