摘要
目的探讨盐酸吡格列酮对兔动脉粥样硬化斑块内基质金属蛋白酶-9(MMP-9)表达的影响及其抗动脉粥样硬化的作用机制。方法新西兰兔36只,随机分为正常对照组、高胆固醇饮食组、高胆固醇饮食加吡咯列酮组。喂养8周后处死每组动物取出从近主动脉弓部到髂动脉分叉处全部主动脉行HE染色、Masson染色及免疫组化染色,观察兔主动脉粥样硬化程度及基质金属蛋白酶-9在各组动脉粥样硬化斑块中的表达。结果高胆固醇饮食加吡咯列酮组动脉硬化程度高于正常对照组,低于高胆固醇饮食组;高胆固醇饮食加吡咯列酮组基质金属蛋白酶-9阳性面积比明显低于高胆固醇饮食组[(39.17±3.52)%vs(49.17±9.12)%,P<0.05],高于正常对照组(P<0.01)。结论吡格列酮具有抗动脉粥样硬化的作用,可使主动脉粥样斑块内基质金属蛋白酶-9表达阳性面积水平下调。
Objective To investigate the effect of piogiltazone on the expression of matrix metalloproteinases-9 in artery of atherosclerosis rabbits and to explore its anti-atherosclerotic mechanism.Methods Thirty-six New Zealand rabbits were randomly divided into 3 groups:control group,high-cholesterol diet group,high-cholesterol diet + pioglitazone group.All animals were fed for 8 weeks and sacrificed.Pathological features of rabbit aortic atherosclerosis were observed by Hematoxylin and Masson staining.Expression of matrix metalloproteinases-9 in aortic atherosclerosis plaque of rabbits was detected by immunohistochemistry.Results The atherosclerosis was more serious in high-cholesterol diet + pioglitazone group than in control group and less than in high-cholesterol diet group.Expression of MMP-9 was lower in high-cholesterol diet + pioglitazone group than that in high-cholesterol diet group [(39.17 ± 3.52) % vs (49.17 ± 9.12) %,P < 0.05] and higher than that in control group (P < 0.01).Conclusion Pioglitazone has anti-atherosclerotic effect by reducing the matrix metalloproteinases-9 expression in aortic atherosclerosis plaque.
