miR-19影响人乳腺癌MCF7细胞增殖、迁移及侵袭的研究

摘要 目的探讨miR-19对人乳腺癌MCF7细胞增殖、迁移及侵袭的影响。方法体外培养人乳腺癌MCF7细胞,分别转染pre-miR-19和miR-19抑制物,然后应用台盼蓝法检测MCF7细胞活性和增殖情况,应用transwell小室法测定MCF7细胞迁移和侵袭能力改变。结果 pre-miR-19转染组的MCF7细胞生长抑制率下降,细胞活性和增殖能力增强,细胞迁移侵袭能力增强;miR-19抑制物转染组的MCF7细胞生长抑制率增高,细胞活性和增殖能力下降,细胞迁移侵袭能力减弱。结论在乳腺癌的发生发展过程中,miR-19发挥原癌基因作用,促进乳腺癌细胞增殖、迁移及侵袭。 Objective To investigate the miR-19 if affect the proliferation, migration ami invasion of human breast cancer MCF7 cell line. Methods After culture of MCF7 cells in vitro,transfeeted with pre-miR-19 anti miR-19 in- hibilor,then the viability and proliferation of MCF7 cells were determined by trypanblue stain,and the migration and invasion of MCF7 cells were detected by tlanswell cell. Results For MCF7 cell transJeeted with pre-miR-19,the cell growth inhibition rate decreased, with the cell viability, proliferation,migration and invasion enhanced. For MCF7 cell transfeeted with miR-19 inhibitors,the cell growth inhibition rate enhanced, with the cell viability,proliferation, migration and invasion decreased. Conclusion The miR-19 have a role of proto-oncogene in human breast cancer de- velopment,and is able to promote proliferation,migration and invasion for breast cancer cells.
