摘要
以文心兰‘小樱桃’花梗腋芽诱导出的丛生芽为增殖培养材料,采用正交设计法,研究基本培养基、6-BA、NAA和水解乳蛋白(LH)4种因素对文心兰丛生芽增殖的影响,以期筛选出各因子的最佳水平,建立文心兰‘小樱桃’丛生芽增殖培养体系。结果表明:各试验因素对文心兰丛生芽增殖影响的主次关系为6-BA>基本培养基>NAA>LH;筛选出文心兰丛生芽增殖的最佳培养基配方为改良1号+6-BA 3.0mg·L-1+NAA0.1mg·L-1+LH 0.5g·L-1,45d平均增殖系数达6.53,有效提高了繁殖效率,为其种苗工程化育苗提供了技术基础。
Peduncle axillary buds of the Little Cherry ,Oncidium ,were induced as explants . The orthogonal design was used to evaluate the effects of the culture medium containing inorganic substances ,6-BA ,NAA and LH on the propagation of Oncidium buds . The result showed that 6-BA ,the base medium ,NAA and LH affected significantly on the bud multiplication . The optimal medium was the Improved Formulation No .1 ,which consisted of the base medium ,6-BA 3.0 mg ? L - 1 ,NAA 0.1 mg ? L - 1 and LH0.5 g ? L - 1 ,and the average propagation coefficient was 6.53 in 45 days . The breeding efficiency was effectively improved . The result appeared to provide a basis for the commercialization of the seedling propagation .
