水稻黑条矮缩病毒(RBSDV)侵染对寄主植物核糖核酸酶基因mRNA表达量的影响 被引量:3

摘要 植物核糖核酸酶能抵抗病原菌侵染。为分析核糖核酸酶在水稻黑条矮缩病毒(RBSDV)侵染过程中的作用,通过灰飞虱人工接种病毒的方法,在寄主植物水稻、小麦和玉米上接种RBSDV,经RT-PCR鉴定病毒成功侵染后,分别以水稻的OsUBQ5、小麦的TaeEF1-α和玉米的ZmaActin作为内参基因,实时荧光定量PCR分析RBSDV侵染后水稻的OsRNS4、小麦的TaeRNS4和玉米的ZmaRNS1基因mRNA表达量的变化。结果显示,感染RBSDV后,寄主植物OsRNS4、TaeRNS4和ZmaRNS1基因的表达量分别是未接种对照植株的3.08倍、1.87倍和3.49倍,表明寄主植物核糖核酸酶基因在RBSDV侵染后被诱导,核糖核酸酶可能在植物抗病毒过程中起作用。 Plant ribonucleases are involved in processes against pathogen attacks. In order to analyze the role of ri- bonucleases in response to rice black-streaked dwarf virus (RBSDV) infection, real-time PCR was used to determine the gene expression level of ribonucleases in the host plant infected with RBSDV. The host plants rice, wheat and maize, were inoculated with Laodelphax striatellus Fallen artificially. RT-PCR analyses confirmed that the host plants were successfully infected with RBSDV. Real-time PCR was employed to evaluate the mRNA expression level of OsRNS4, TaeRNS4 and Zm- aRNS1 in RBSDV-infected rice, wheat and maize, respectively. OsUBQ5 of rice, TaeEF1-α of wheat, ZmaActin of maize were taken as reference genes. The results showed that RBSDV infection induced the expression of ribonucleases in plants, and the mRNA expression levels of OsRNS4, TaeRNS4 and ZmaRNS1 were 3.08 times, 1.87 times and 3.49 times as much as the non-infected plants, which indicated that plant ribonucleases might play important roles in acting against virus infection.
