摘要
目的:研究蕲艾提取液药物血清对大鼠肝星状细胞(HSC)凋亡的影响。方法:传代培养的HSC-T6与蕲艾提取液药物血清共同培养24小时后,采用Annexin V/PI对双染结合流式细胞仪检测细胞凋亡。结果:蕲艾提取液药物血清组早期细胞凋亡率显著高于空白对照组及生理盐水血清组(P<0.01),并且随着血清浓度的增加,HSCT6早期细胞凋亡率逐渐增高,呈剂量依赖关系。结论:蕲艾提取液药物血清促进HSC凋亡,这可能是蕲艾提取液药物血清抗肝纤维化作用机制之一。
Objective: To investigate the effect of artemisia argyi extract solution drug serum on apoptosis of hepatic stellate cells (HSC) in rat. Methods: HSC-T6 was incubated with different concentrations of artemisia argyi extract solution drug serum (5%, 10%, 20% ) for 24 hours, the apoptosis of HSC-T6 was detected by flow cytometry with Annexin V and PI staining. Results: The percentage of early apoptotic cells of artemisia argyi extract solution drug serum group was significantly higher than blank control group and saline group (P 〈 0.01 ) . With the increase of serum concentration, HSC-T6 cell early apoptosis rate was gradually increased in a dose-dependent manner. Conclusion: Artemisia argyi extract solution drug serum can promote HSC apoptosis, which may be one of the mechanisms of anti-liver fibrosis.
