越南伯克霍尔德氏菌B418培养条件的优化 被引量:2

摘要 以越南伯克霍尔德氏菌B418为研究对象,采用均匀设计实验、正交设计实验和单因子实验研究了各种因素对菌株生长的影响.确定了B418生长最佳培养基配方为(g· L-1):葡萄糖10,蛋白胨5,硫酸镁2,硫酸铵3,玉米粉30,黄豆粉10,磷酸二氢钾1,磷酸氢二钾1,初始pH值7.5.主要影响因子为硫酸镁,其次为硫酸铵.其它优化培养条件包括:接种量5%、培养温度28℃、培养时间28 h、转速180 r·min-1.利用优化条件上500 L发酵罐,通气量1 ~3 L· min-1,B418在16 h进入对数生长期,28 h进入稳定期,活菌数达210.5×108 cfu·mL-1. Studied the influence of different factors on the growth of Burkholderia viemamiensis 13418 by using uniform design experimentation, orthogonal experimentation and simple factor design of experiment, the optimum medium component were including (g · L-1 ) : glucose 10, peptone 5, MgSO4 2, (NH4)2SO4 3, corn meal 30, soybean meal 10, KH2P04 1, K2 HPO4 1, initial pH 7. 5. The main factors affecting bacterial population were MgSO4 and ( NH4 ) 2 S04- Other culture condition optimization were including inoculation quantity 5%, culture temperature 28℃, culture time 28 h, rotate speed 180 r · min- 1 After culture in a 500 L fermentor using optimum condition, the air flux were 1 · 3 L · miu- 1, the strain was into logarithmic growth phase after 16 h, and into stationary phase after 28 h. The bacterial population were to reach 210. 5 x 10s cfu · mL-1 in the end.
