摘要
目的优选山茱萸中多糖提取工艺,并建立山茱萸多糖含量的测定方法。方法采用水提醇沉法提取山茱萸中多糖,以料液比、提取时间和提取温度为考察因素,以多糖提取率为评价指标,采用正交试验优选提取工艺;应用苯酚-硫酸法测定山茱萸多糖含量,以苯酚用量、浓硫酸用量和反应时间为考察因素,采用正交试验优选显色条件。结果山茱萸中多糖最佳提取工艺为料液比1∶16、提取时间1.5 h、提取温度80℃;用5%苯酚1.0 ml,浓硫酸6 ml,反应时间30 min进行显色。结论正交设计优化山茱萸多糖的提取条件,多糖得率高;苯酚-硫酸法测定山茱萸多糖含量稳定性、回收率较好。
Objective To optimize the extraction process for polysaccharides and establish the method for determination of the content of polysaecharides from Fructus Corni. Methods Aqueous extraction was used for the extraction of Fructus Comi polysac- charities. The extraction technology was optimized by orthogonal test with solid - liquid rate , extraction time and extraction temper- ature as factors using extraction rate as index. The content of Fructus Corni polysaccharide was determined by phenolvitriolic color- imetry. With the application of the orthogonal test with the amount of phenol, sulfuric acid and reaction time as factors, the best col- oured conditions were selected. Results The optimum process conditions with aqueous extraction were as follows : solid - liquid ra-tio of 1:16, extraction time of 1.5 h, extraction temperature of 80℃. The best coloured conditions were as follows :5% phenol of 1. 0 ml, sulfuric acid of 6 ml, reaction time of 30min. Conclusion The orthogonal test is helpful to optimize extraction conditions and coloured conditions of Fructus Corni polysaccharide. The extraction rate of polysaccharide is high. Phenol - vitriolic colorimetry was a stable and reproducible method to determine content of polysaccharide in Fructus Corni.
