当归补血汤调控骨髓造血机理及对造血微环境的影响 被引量:39

摘要 目的探讨当归补血汤调控骨髓造血的机理。方法 50只ICR小鼠随机分为5组,每组10只。除正常组外其他组小鼠连续腹腔注射多柔比星4 mg·kg^(-1)·d^(-1),共3 d,正常组给予同等体积的生理盐水。注射多柔比星3 d后,当归补血汤低、中、高剂量组分别连续腹腔注射补血汤5、15、25 g·kg^(-1)·d^(-1),共7 d,正常组和模型组给予同等体积的生理盐水。给药结束制备骨髓单细胞悬液,体外培养观察骨髓基质细胞生长状态,流式细胞仪检测骨髓细胞增殖和黏附情况。结果当归补血汤组骨髓基质细胞生长状态好于正常组和模型组,骨髓细胞之间的聚集明显。模型组聚集细胞数占总细胞比例为(4.00±1.10)%,低于正常组[(3.07±2.35)%],但无显著差异(P>0.05)。当归补血汤低、中、高剂量组聚集细胞数目占总细胞数比例分别为(5.40±2.91)%、(7.36±2.32)%和(9.27±3.35)%,均高于模型组(P<0.05或P<0.01)。正常组G2期细胞比例高于其他四组(P<0.05),当归补血汤各剂量组G2/M期细胞比例与模型组比较无显著差异(P>0.05)。结论当归补血汤可能通过改善基质细胞的状态、增强骨髓细胞之间的黏附来改善骨髓造血微环境。 AIM To study the mechanism of Dangguibuxuetang regulating bone marrow hematopoietic METHODS Fifty ICR mice were randomly divided into 5 groups (n = 10), including normal group, modelgroup and Dangguibuxuetang groups which had three different doses. Mice received either doxorubicin (4 mg. kg^-1 .d^-1, model group) or the equal volume of saline (normal group) daily via intraperitoneal injection for 3 days. For Dangguibuxuetang groups, mice received three different dosages of Dangguibuxuetang daily via intraperitoneal injection for 7 days after 3 days doxorubicin administration, respectively, including low- dose (5 g^-1.d^-1), medium-dose (15 g^-1.d^-1) and high-dose (25 g^-1.d^-1) group. Mice in normal and model groups received matching vehicle (saline) injection. Bone marrow cell suspension was collected to examine bone marrow stromal cell viability. Bone marrow cell proliferation and adhesion ability were measured by flow eytometry. RESULTS Compared with the normal group and model group, Dangguibuxuetang treatment accelerated the growth of bone marrow stromal cells and enhanced bone marrow cell aggregation. The aggregate cell numbers in model group ((4.00 ± 1.10) %) was lower than the normal group ((3.07 ± 2.35) %), but with no significant difference (P 〉 0.05 ). The aggregate cell numbers in all three Dangguibuxuetang groups were higher than that in model group (P 〈 0.05 or P 〈 0.01 ). The proportion of cells in G2 phase in the normal group was the highest among all groups (P 〈 0.05). No significant difference in the proportion of cells in G2/M phase was observed between Dangguibuxuetang group and model group (P 〉 0.05) . CONCLUSION Dangguibuxuetang improves bone marrow microenvironment by accelerating the growth of stromal cells and enhancing the adhesion between bone marrow cells.
