摘要
目的研究阿司匹林诱导宫颈瘤Hela细胞凋亡的机制。方法培养人宫颈癌Hela细胞,加入不同浓度阿司匹林(1.0、5.0、10.0 mmol/L)干预培养。应用流式细胞技术检测Hela细胞凋亡和细胞周期的变化;用RT-PCR检测细胞凋亡相关基因bcl-2及bax转录水平的改变。结果与对照组(4.13%±0.76%)相比,阿司匹林干预组Hela细胞凋亡率(11.71%±0.86%,18.15%±0.46%,30.81%±0.46%)明显增高(P<0.05);随着阿司匹林浓度增加,G0/G1期细胞比例下降,G2/M期细胞比例升高(P<0.05);阿司匹林干预培养后Hela细胞中bcl-2基因mRNA表达明显降低,而bax基因mRNA表达升高明显,两种基因表达较对照组均有统计学意义(P<0.05)。结论阿司匹林能诱导宫颈癌Hela细胞凋亡,其机制与抑制bcl-2表达,上调bax表达有关。
Objective To explore the apoptosis effect and mechanism of aspirin on human cervical cancer hela cell line.Methods Hela cell was cultured and induced with aspirin of concentration gradient.The effect of aspirin on hela cell apoptosis and cell cycle was observed by cell flow cytometry(FCM).The levels of bcl-2 and bax mRNA were detected by RT-PCR.Results Compared with control group,apoptosis percentage of Hela cells treated with various-dose aspirin had improved significantly(P < 0.05) ; Hela cells in G0/G1 stage of were decreased,and in G2/M stage were increased ; aspirin might down-regulate the expression of bcl-2 mRNA,up-regulate the expression of bax mRNA.Conclusions The apoptosis mechanism of aspirin on human cervical cancer Hela cell line involves decreasing the bcl-2 expression and enhancing the expression of bax.
