摘要 目的探讨糖尿病大鼠肾脏中Toll样受体4(TLR4)的表达及与巨噬细胞浸润的关系。方法 STZ注射制备糖尿病大鼠模型。用免疫组织化学法检测大鼠肾脏巨噬细胞的浸润及TLR4的表达。用Western blot及RT-PCR方法检测大鼠肾脏TLR4蛋白及mRNA的表达。结果成模后,TLR4在肾小管的表达增加。DM组大鼠2、4、8周TLR4/β-actin表达高于NC组。DM组2、4、8周TLR4/GAPDH mRNA表达均高于NC组。DM组4、8周肾间质巨噬细胞浸润数均高于NC组。结论在糖尿病大鼠肾脏中,TLR4表达上调,与炎症浸润相关。 Objective To investigate whether the Toll-like receptor 4 (TLR4) signaling is activated in diabetic nephropathy. And to investigate the relationship between TLR4 expression and renal macrophage infiltration. Methods Diabetes was induced by a single intravenous injection of streptozotocin. Immunohistochemical method was used for measuring macrophage infiltration and TLR4 expression. Western Blot was used for measuring TLR4 protein. RT-PCR was used for measuring TLR4 mRNA expression. Results In diabetic rats, there was a significant up-regulation of TLR4 protein and mRNA expression in the kidney. In the DiM group, the expression of renal TLR4/β-actin in the 2^nd, 4^th, and 8^th weeks was higher than in the NC group; the expression of renal TLR4/GAPDH mRNA in the 2^nd, 4^th, and ^th weeks was higher than in the NC group; and the renal macrophage infiltration in the kidney in the 4^th and 8^th weeks was more than in the NC group. Conclusion Enhanced renal expression of TLR4 is associated with inflammatory infiltration in diabetic nephropathy.
