摘要
目的:探讨姜黄素对人乳腺癌细胞株(MCF-7)中miR-21表达的影响及其意义。方法:采用浓度分别为2.5μmol/L,5μmol/L,10μmol/L,20μmol/L的姜黄素处理MCF-7细胞株,采用实时荧光定量逆转录聚合酶链反应(Realtime PCR)的方法检测其miR-21的表达。免疫印迹法(Western blot)方法检测miR-21调控的目的蛋白PDCD4的表达。结果:随着姜黄素浓度的升高,miR-21的表达量明显下降,目的蛋白PDCD4的表达量明显上升。结论:在乳腺癌细胞中,姜黄素通过调控miR-21及其目的蛋白的表达而发挥明显抑制肿瘤细胞增殖的作用。
Objective: To explore the effect of curcumin on miR -21 level in human breast cancer MCF -7 cell line and its signif- icance. Methods: Curcumin of different concentrations (2. 5 μmol/L, 5μmoL/L, 10μ mol/L, 20 μmol/L) were respectively used to treat MCF -7 cell line, real -time fluorescent quantitative PCR was used to detect the expression of miR -21. Western blotting was used to de- tect the expression of target protein PDCIM regulated by miR -21. Results: The expression level of miR -21 decreased significantly with the increase of curcumin concentration, and the expression level of PDCD4 increased significantly. Conclusion : Curcumin can obviously in- hibit the vroliferation of breast cancer cells by reaulating the ext3ression levels of miR - 21 and its target protein.
