商陆醋炙前后化学成分和药效比较 被引量:10

Comparison of Chemical Composition and Efficacy before and after Processed Phytolaccae Radix with Vinegar

摘要 目的:比较商陆醋炙前后化学成分和药效的变化情况。方法:按2010年版《中国药典》商陆项下醋炙法制备醋商陆。采用硫酸-香草醛比色法测定总皂苷含量;HPLC—ELSD测定商陆皂苷甲含量,色谱条件为Lichrospher—C18色谱柱(4.6mm×250mm,5Ixm),流动相甲醇-0.4%乙酸(65:35),流速1.0mL·min^-1,柱温40℃,进样量20μL,ELSD条件为漂移管温度90℃,载气流速2.5L·min^-1。SD大鼠随机分成生理盐水组、阳性药组(氢氯噻嗪)、生商陆组和醋商陆组,以生理盐水形成盐水负荷模型,用大鼠代谢笼法观察大鼠0~5h尿量;ICR小鼠随机分成生理盐水组、阳性药组(芒硝)、生商陆组和醋商陆组,采用墨汁推进法比较生商陆和醋商陆对小鼠小肠推进率的影响。结果:商陆经醋炙后总皂苷质量分数由1.66%升至1.91%,商陆皂苷甲质量分数由0.64%增至0.82%,大鼠排尿量增多,小鼠小肠推动作用减弱。结论:商陆醋炙后总皂苷和商陆皂苷甲含量均升高,泻下作用缓和,长于利尿逐水。 Objective: To compare changes of chemical composition and efficacy before and after Phytolaceae Radix processing with vinegar. Method: Phytolaccae Radix was processed by according to vinegar processing method in the 2010 edition of ' Chinese Pharmacopoeia'. The content of total saponins was dtermined by sulfuric acid-vanillin eolorimetry; The content of esculentoside A was determined by HPLC-ELSD, chromatographic conditions were: Lichrospher-C18 column (4.6 mm x 250 mm, 5 μm), mobile phase of methanol-0.4% acetic acid (65:35), flow rate 1.0 mL-min-X, column temperature 40 ℃, injection volume 20 μL;ELSD conditions were: drift tube temperature 90 ℃ , flow rate of carrier gas 2.5 L ·min-l SD rats were randomly divided into normal saline group, positive drug group (hydrochlorothiazide), crude and processed Phytolaccae Radix group, brine loading model formed with saline, then we observed urine of rats within 5 hours by metabolic cage method. ICR mice were randomly divided into normal saline group, positive drug group (mirabilite) , crude and processed Phytolaceae Radix group, then we compared small intestinal propulsive rate between crude and processed Phytolaccae Radix by ink propulsion method. Result : After processing of Phytolaccae Radix with vinegar, the content of total saponins increased from 1.66% to 1.91% , the content of esculentoside A increased from 0.64% to 0.82% , diuretic efficacy on rats enhanced and purgative efficacy on mice moderated. Conclusion : After being processed with vinegar, the content of total saponins and esculentoside A both increased, diuretic efficacy enhanced and purgative efficacy moderated.
