抗骨增生片对兔膝骨关节炎软骨中白细胞介素-1β、转化生长因子-β1表达的影响 被引量:2

摘要 目的观察抗骨增生片对兔膝骨关节炎软骨中白细胞介素-1β(IL-1β)、转化生长因子-β1(TGF-β1)表达的影响,以探讨抗骨增生片治疗骨关节炎可能的作用机制。方法将30只健康成年雄性日本大耳白兔随机分成3组,即正常对照组、模型组、抗骨增生片组,每组10只。模型组、抗骨增生片组于右膝内注射木瓜蛋白酶生理盐水溶液建立膝骨关节炎模型。抗骨增生片组按成人的每日每kg体质量用药量的6倍剂量给药,溶于0.9%氯化钠注射液10 mL中灌胃,每日1次。正常对照组、模型组用等量0.9%氯化钠注射液灌胃。共4周。免疫组化法检测各组兔膝关节软骨中IL-1β、TGF-β1的表达情况。结果模型组软骨IL-1β阳性表达率较正常组增高(P<0.01),抗骨增生片组IL-1β阳性表达率较模型组均降低(P<0.01);模型组软骨TGF-β1阳性表达率较正常组降低(P<0.01),抗骨增生片组TGF-β1阳性表达率较模型组增高(P<0.01)。结论抗骨增生片可能通过增加TGF-β1的合成,进而抑制IL-1β的表达,从而促进了软骨修复,保护了关节软骨,起到治疗骨性关节炎的作用。 Objective To observe the effect of Kangguzengsheng tablet on the expression of IL - Iβ,TGF - β1 in cartilage of rabbits knee osteoarthritis and elucidate the potential mechanism of Kangguzengsheng tablet on oste- oarthritis. Methods 30 healthy male rabbits ( weight 2.5 kg + 150 g) were randomly divided into three groups : A, normal control group, B, model group, C, Kangguzengsheng Tablet group. The osteoarthritis models of rabbits were established by injecting papain to the right knees of experimental rabbits in group B and C. Each rabbit of group C was intragastricly administrated Kangguzengsheng Tablet (dissolved in lOml normal saline) for four weeks, according to its equivalent dosage/kg/day (6 times of person' s dosage/kg/day), 1/day; rabbits of group A and B were given equivalent normal saline for 4 weeks. The expression of IL - 1 β, TGF - β1 in cartilage of rabbit' s knee was measured by immunohistochemistry. Results The positive expression rate of IL - 1 β in cartilage of group B was higher than that of group A ( P 〈 0. O1 ). The positive expression rate of IL - 1 β in cartilage of group C was lower than that of group B ( P 〈0.01 ). The positive expression rate of TGF - β1 in cartilage of group B was lower than that of group A ( P 〈 0.01 ). The positive expression rate of TGF- β1 in cartilage of group C was higher than that of group B (P 〈0. 01 ). Conclusion By means of increasing the TGF - β1 synthesis, Kangguzengsheng tablet may inhibit IL - 1 beta expression, so as to promote the cartilage repair, protect the articular cartilage, to benefit the treatment of osteoarthritis.
