摘要
目的:探讨灭多威对家蝇胚胎细胞系MDEC-07114细胞凋亡的影响。方法:1、实验分组:不同浓度灭多威组(1 mg/mL组、2 mg/mL组、4 mg/mL组、8 mg/mL组),同时设正常组、1%丙酮组;2、HE染色法观察MDEC-07114细胞凋亡形态,并测定细胞凋亡指数;3、TUNEL检测MDEC-07114细胞凋亡。结果:1、不同浓度灭多威作用于MDEC-07114后,从2 mg/mL组开始见典型凋亡细胞,细胞凋亡指数增加,且随浓度的递增而逐渐升高;2、TUNEL检测:2 mg/mL组出现明显的绿色荧光,4 mg/mL组有所增加,8 mg/mL组荧光最强。结论:灭多威可诱导MDEC-07114细胞凋亡。
Objective of this study was to explore effect of Methomyl on apoptosis of Musca domestica embryonic cell line (MDEC-07114). Experimental groups were the different concentrations of Methomyl groups (1 mg/ml, 2mg/ml, 4mg/ml and 8mg/ml), normal group and 1% acetone group. The morphology of apoptosis cells was observed by hematoxylin-eosin staining, and the apoptosis index (A1) was measured. The apoptosis of MDEC- 07114 cell was detected by TUNEL Results showed that from the 2mg/ml groups of Methomyl, the typical apoptotie cell began to appear. Moreover, with the increase of the concentration, the apoptosis index (AI) gradually increased. TUNEL assay showed that at the 2mg/ml groups, MDEC-07114 ceils appeared visible green fluorescence, the green fluorescence in ceils enhanced at the 4mg/ml groups, and it was the strongest at the 8mg/ml groups. Conclusion of this experiment was methomyl could induce apoptosis ofMDEC-07114 cells.
