摘要 目的 研制ABO反定型免疫胶体金试剂,以解决反定型细胞需低温保存、保存期短的问题.方法 提取O、A和B型红细胞膜抗原并应用胶体金标记,研发ABO反定型免疫胶体金试剂.研究该试剂的最佳反应条件.用ABO反定型免疫胶体金试剂检测已知血型的临床全血标本200例,并与传统人ABO血型标准反定型细胞比较,评估其反应的特异性、灵敏度和准确性.结果 结果显示10μl金标记红细胞膜抗原和45μl左右的检测血清/血浆有最佳的凝结强度,最佳震荡反应转速为80 ~90 r/min,反应最佳时间为8~12 min.与传统人ABO血型标准反定型细胞相比,ABO反定型免疫胶体金试剂的准确性、特异性均为100%,灵敏度不低于反定型红细胞.结论 ABO反定型免疫胶体金试剂可以用于常规血型鉴定. Objective To develop immunity colloidal gold reagent for ABO reverse typing so as to solve the problems of preservation at a low temperature and short shelf life. Methods Type O, A and B membrane antigens of red blood cells were ex- tracted and labeled with colloidal gold, which were used as immunity colloidal gold reagent for ABO reverse typing. The optimum re- action conditions of the immunity colloidal gold reagent were determined. A total of 200 blood samples of known blood types were de- tected with immunity colloidal gold reagent, and compared with traditional reagent cells. The specificity, sensitivity and accuracy of the immunity colloidal gold reagent were assessed. Results The optimum agglutination intensity existed in 10 μl of colloidal gold- labeled membrane antigens and 45 μl of serum (or plasma) was generated. The optimum oscillation speed was 80-90 r/min, and the optimum reaction time was 8-12 min. The specificity and accuracy of this immunity colloidal gold reagent was 100% , and the sensi- tivity was not lower than that with traditional reagent red cells in reverse ABO typing. Conclusion The immunity colloidal gold rea- gent for ABO reverse typing can be used in routine blood group examination.
