摘要
目的探讨Notch1及其信号通路相关基因在乙型肝炎感染过程中的表达及临床意义。方法选取急性乙型肝炎(AHB)患者21例和慢性乙型肝炎(CHB)患者25例,正常对照20例,抽取抗凝外周血,以免疫磁珠法分离CD4+T细胞和CD8+T细胞,分别抽提两种细胞的总RNA,荧光定量PCR分析Notch1、Hes1、Jag1和NF-κB的mRNA表达情况。结果 CD4+T细胞中仅NF-κB在AHB和CHB组的表达均较对照组显著增加(P<0.01),而其他基因各组间表达无统计学意义(P>0.05);CD8+T细胞中Notch1、Jag1和Hes1的mRNA表达在AHB组显著高于其他两组(P<0.01),同时其在CHB组表达高于对照组,差异有统计学意义(P<0.01),而NF-κB表达无统计学差异(P>0.05)。结论 Notch1相关基因的异常表达可能通过调节CD4+和CD8+T细胞的异常分化参与调节乙肝病毒感染。
Objective To investigate the expression of Notchl and its related genes in HBV infection and its clinical significance. Methods A total of 21 patients with acute hepatitis B (AHB), 25 patients with chronic hepatitis B (CHB) and 20 normal con-trols were involved in this study. CD4^+ and CD8^ + T cells were isolated from peripheral blood. Total RNA was extracted. Notchl, Hesl, Jagl and NF-KB were analyzed by fluorescence quantitative PCR. Results The expression of NF-κB in CD4^ + T cells in the patients with AHB and CHB was higher than that in the normal controls (P 〈 0.01 ). No statistic significance was found for the oth-er genes (P 〉 0.05). The mRNA expression of Notchl, Jagl and Hesl of CD8^+ T cells in the patients with AHB was higher than thatin the other two groups(P 〈0.01 ), and the expression in the patients with CHB was higher than that in the normal controls (P 〈 0.01 ). Conclusion Abnormal expression of Notehl and its related genes may regulate HBV infection by controlling the differentia-tion of CD4^ + and CD8^ + T ceils.
