埃索美拉唑对幽门螺杆菌诱导胃上皮细胞表达细胞因子的影响

Effect and Molecular Mechanism of Esomeprazole on Helicobacter pylori-induced Cytokines Production in Gastric Epithelial Cells

摘要 目的探讨埃索美拉唑对幽门螺杆菌(Hp)感染的胃上皮细胞的体外抗炎效应。方法体外培养胃上皮细胞系AGS,加入不同浓度埃索美拉唑和培养的Hp感染8或24 h。Western blot检测和实时定量PCR分别检测AGS细胞中红素氧合酶1(HO-1)蛋白和mRNA表达情况;ELISA检测白细胞介素-1β(IL-1β)和IL-8的产生。同时采用RNA干扰HO-1表达后,观察其对Hp诱导IL-1β和IL-8产生的影响。结果埃索美拉唑孵育8 h后,能显著抑制Hp诱导的IL-1β和IL-8表达。同时,埃索美拉唑能在转录翻译水平分别调控HO-1 mRNA和蛋白表达。HO-1 siRNA沉默HO-1表达后,与Hp组相比,IL-1β和IL-6分泌进一步增高。结论埃索美拉唑可能通过上调HO-1的表达而发挥对胃上皮细胞的抗炎作用。 ObjectiveTo investigate the anti-inflammatory effects of Esomeprazole in H. pylori-infected gastric epithe- lial cells in vitro. Methods Gastric epithelial cell line AGS was cultured in vitro, and was incubated with Esomeprazole and H. pylori for 8 or 24h. Expression of Heine oxygenase-1 mRNA and protein was detected by Western blot and real-time PCR,respectively;production of IL-6 and IL-8 were detected by ELISA. RNA interference was used to silent HO-1 expres- sion, and H. pylori-induced production of IL-1β and IL-8 were detected. Results After incubated for 8 h, esomeprazole could significantly inhibit H. pylori induced IL-1β and IL-8 production. And esomeprazole could also induce AGS cells ex- pression of HO-1 mRNA at the transcriptional and translational level. Furthermore, silencing of HO-1 expression could in- crease AGS cell secretion of IL-1 β and IL-8, as compared with that in H. pylori group. Conclusion HO-1 is a target of esomeprazole in gastric epithelial cells. HO-1 induction might account for the gastroprotective effects in vasoprotection.
