摘要
目的 探索生产降纤酶的合适工艺 ,改进目前降纤酶生产过程中试剂用量大 ,工艺繁琐 ,不易掌握 ,收率低 ,质量不稳定的缺陷。 方法 采用二步梯度洗脱 ,改变 Tris缓冲液 p H值 ,洗脱液的盐浓度及洗脱速度。改进 DEAE-sephadex A50的平衡方法 ,精简 DEAE-cellulose DE52工序。 结果 分离出 9个峰 ,平均收率提高到 70 0支 /g,生产周期 6天 ,比原工艺缩短 5天 ,减少了试剂用量 ,找到去除热原的方法。 结论 与原工艺相比 ,新方法明显提高了经济效益 。
Objective There exits several shortcomings in the present procedure of defibrase production:Large amount of reagent consumed,overelaborate technology to handle,low rate of returns and unsteady quality.It is our hope in this study to find out a new procedure more suitable for production,which can achieve greater,faster,better and more economical results. Methods The DEAE-cellulose D52 procedure is expected to be simplified by adopting the two-step gradient elution,changing the pH value of Tris buffer,the salt concentration of elute and the rate of elution,and improving the balance of DEAE-sephadex A50. Results The inproved procedure is expected to increase the separated peaks from five to nine,raise the average rate of returns from 550 to 700 ampoles/g, shorten the cycle of production from eleven to six days,reduce the amount of reagent,and find a method of disgarding pyrogen. Conclution The new technology can greatly increase economic results and is a practicable production procedure as compared with the original one . [
