酶法制备低聚木糖中木二糖的提纯与色谱鉴定

Separation and purification of xylobiose from xylooligosaccharides and HPLC determination

半纤维素经木聚糖酶水解得到混合的低聚木糖 ,用葡聚糖凝胶柱SephadexG - 1 0层析分离后 ,出现四个组分峰 ,经高效液相色谱分析鉴定 ,其中 3号峰为木二糖纯组分峰。结果表明 :利用2 .5cm× 1 2 0cm凝胶柱 ,以蒸馏水为流动相 ,流速为1 0ml/h ,在室温下 。

The separation and purification of xylooligosaccharides from hydrolysate of hemicellulose by xylanases were studied. The result showed that xylooligosaccharides could be effectively isolated by the Sephadex G-10 column and the pure samples of xylobiose were obtained. This result was fully confirmed by HPLC analysis. The separating conditions were as follows: column size, 2.5 cm×120 cm; mobile phase, water; flow rate, 10ml/h; column temperature, 30℃.
