摘要
在 p H3.0时 ,去除 85和 95~ 10 5USP u/ mg粗品肝素钠的杂蛋白分别使用0 .0 5%和 0 .0 3%的 YNB99- 1。 12 0 USP u/ mg以上的粗品在 p H中性下直接使用0 .0 5%的 YNB99- 1即可。在氧化精制中 ,加入 0 .0 0 5%的 YNB99- 1可使 H2 O2 用量减少至 1.5% ,氧化时间缩短至 12 h。精品效价 170 USP u/ mg以上 ,产品收率 90 %以上。
Precipitator YNB 99 1 can remove acidic protein in crude heparin sodium under the conditions of pH 3.0 and 0.05% YNB 99 1 which is for the crude of potency 85 USP u/mg,as well as pH 7.0 and 0.03% the precipitator for that of 95 to 105 USP u/mg.To the purification by oxidization method,either H 2O 2 quantity is decreased to 1.5%(48 hours) or oxidization time shorted to 12 hours(3.0% H 2O 2) with adds of 0.005% YNB 99 1 separately.The quality of purified product,especially absorption of 260 nm and 280 nm,is better than the traditional processs.The collecting ratio of the purified product is more than 90% and its potency higher than 170 USP u/mg.
