摘要
目的观察和研究人工合成家蝇抗真菌肽MAF-1A对白色念珠菌致病性的影响。方法以白色念珠菌ATCC10231为效应菌,检测MAF-1A在MIC浓度下不同时间白色念珠菌的孢子萌发率及芽管形成率,瑞氏染色后显微镜计数,观察MAF-1A预处理后及干预下,酵母相和菌丝相白色念珠菌定植人口腔黏膜上皮细胞能力变化;分别设立氟康唑(fluconazole,FLC)和PBS为阳性对照和阴性对照。结果 MAF-1A作用白色念珠菌后,孢子萌发率及芽管形成率小于阳性对照和阴性对照组(P<0.05);MAF-1A可减少白色念珠菌酵母相细胞或菌丝相细胞对人口腔黏膜上皮细胞的粘附数,且减少酵母相细胞粘附作用强于菌丝相细胞(P<0.05);MAF-1A预处理白色念珠菌后,对人口腔黏膜上皮细胞粘附数少于MAF-1A干预组的细胞粘附数(P<0.05)。结论 MAF-1A可通过抑制白色念珠菌孢子萌发和芽管的形成以及有效降低白色念珠菌对人口腔黏膜上皮细胞粘附能力,从而影响白色念珠菌的致病能力。
Objective To probe the effect of synthetic Musca domestica antifungal peptide-1A (MAF-1A) on the pathogenicity of Candida albicans. Methods Blastospore germination and germ tube formation of Candida albicans ATCC10231 were detected and fungal numbers of adherence to human buccal epithelial cells (BEC) in vitro were counted for yeast form and hyphal form of Candida albicans respectively using Wright’s staining after exposing to MIC concentration of MAF-1A, with fluconazole as positive control and PBS as negative control. Results Comparing with the controls, MAF-1A significantly decreased the blastospore germination and germ tube formation of Candida albicans (P〈0.05), and reduced the numbers of both yeast form and hyphal form cells adhered to BEC, showing stronger inhibition effects on the yeast form cells than the hyphal form cells. The number of Candida albicans adhered to BEC in the MAF-1A pretreatment group was less than that in the MAF-1A intervention group (P〈0.05). Conclusions MAF-1A affects the pathogenicity of Candida albicans by inhibiting blastospore germination, germ tube formation and the adhesion ability to BEC.
