红花黄素体外诱导BMSCs分化为神经元样细胞的实验研究 被引量:3

摘要 目的探讨红花黄素在体外诱导大鼠骨髓间充质干细胞(BMSCs)转变为神经元样细胞的效果。方法2012年8月至2013年4月,采用全骨髓贴壁培养法培养大鼠BMSCs,取P3代BMSCs接种培养板,分3组,每组6孔:A组(红花黄素组)和B组(对照组1)待细胞接近50%融合后,每孔内加入10μg/L的bFGF预诱导24h,PBS洗涤2次后,A组再加入含红花黄素的无血清DMEM诱导液诱导24h后弃掉诱导液,加入DMEM培养基继续培养;B组直接加入DMEM培养基继续培养;C组(对照组2)始终用含15%DMEM培养基培养。各组于倒置相差显微镜下观察细胞形态学变化。各组待细胞达90%融合时,采用免疫荧光法检测胶质纤维酸性蛋白(GFAP)、神经丝蛋白M(NFM)、神经元特异性烯醇化酶(NSE)的表达来鉴定分化后细胞。结果A组BMSCs呈渐进性地向神经元样细胞转化;NSE、NFM染色呈强阳性:NSE阳性细胞率为(63.36±1.26)%;NFM阳性细胞率(56.19±3.64)%;B、C组为阴性;3组GFAP表达均为阴性。结论红花黄素在体外诱导大鼠BMSCs可分化为神经元样细胞。
