摘要
目的:建立肺癌恶病质模型,探讨保元解毒汤对模型动物生活质量的干预作用。方法:用Lewis肺癌细胞株皮下接种C57BL/6小鼠,建立肺癌恶病质小鼠模型。40只小鼠随机分为正常对照组(NC)、模型对照组(MC)、中药干预组(TCM)和醋酸甲羟孕酮组(MPA),每组各10只。观察小鼠体质量、摄食量、瘤体积、瘤质量、腓肠肌质量和生存期等生活质量相关指标变化。结果:在接种肿瘤细胞后第1、8、18和29天分析小鼠体质量及摄食量变化有差异,尤其以第29天差异最为显著。MC组小鼠体质量为(20.02±0.45)g,TCM组小鼠体质量为(23.75±0.21)g,MPA组小鼠体质量为(23.32±0.44)g,TCM组较MC组体质量明显增加,P=0.009;TCM组较MPA组体质量差异无统计学意义,P=0.251。MC组小鼠摄食量为(1.13±0.15)g,TCM组小鼠摄食量为(2.26±0.57)g,MPA组小鼠摄食量为(2.50±0.15)g,TCM组较MC组摄食量明显增加,P=0.001;TCM组较MPA组摄食量差异无统计学意义,P=0.473。在接种肿瘤细胞后第1、8、18和29天分析小鼠体质量及摄食量变化有差异,尤其以第29天差异最为显著。药物治疗后,MC组小鼠腓肠肌质量为(79.31±1.73)mg,TCM组小鼠腓肠肌质量为(107.70±1.57)mg,MPA组小鼠腓肠肌质量为(82.48±1.52)mg,TCM组较MC组明显增加,P=0.005;较MPA组明显增加,P=0.007。接种29d后,MC组小鼠瘤体积为(7 664.34±286.00)mm3,TCM组小鼠瘤体积为(3 935.44±561.40)mm3,MPA组小鼠瘤体积为(5 960.22±290.00)mm3,TCM组较MC组明显缩小,P=0.005;较MPA组明显缩小,P=0.007;MC组小鼠瘤质量为(8.06±1.82)g,TCM组小鼠瘤质量为(3.78±0.98)g,MPA组小鼠瘤质量为(6.65±0.79)g,TCM组较MC组明显缩小,P=0.043;较MPA组明显缩小,P=0.052;MC组小鼠生存期为(29.90±3.21)d,TCM小鼠生存期为(34.80±3.39)d,MPA小鼠生存期为(30.30±3.20)d,TCM组较MC组明显延长,P=0.043;较MPA组明显延长,P=0.045,MC组与MPA组生存期差异无统计学意义,P=0.307。结论:保元解毒汤对改善肺癌恶病质小鼠生活质量有较好作用,可以延长其生存期。
OBJECTIVE To establish the cancer cachexia model and to explore the intervention effect of Bao Yuan detoxofication soup in improving the quality of life of cancer cachexia model animal. METHODS: Mice model of cancer ca chexia was established by inoculating Lewis lung cancer cell lines to C57BL/6 mice subcutaneously. Forty mice were ran domly divided into 4 groups:normal control group,model control group,TCM group and MPA group, with 10 mice in each group. The changes of quality life conditions (body weight, food intake, tumor volumes, tumor weight, gastrocnemius mus cle weight and survival time) were observed in the experiment. RESULTS: Body weight:the body weight of MC group was (20. 02±0.45) g;TCM group was (23.75±0.21) g;MPA group was (23. 32±0.44) g. The body weight of TCM group was significantly higher than that of the MC group(P= 0. 009), which was no difference compared with MPA group. Theconsumption of feed:the consumption of feed of MC group was (1.13±0.15) g;TCM group was (2.26±0.57) g;MPA group was (2.50±0. 15) g. The consumption of feed of TCM group was significantly higher than that of the MC group (P= 0. 001), having no difference compared with MPA group. Muscle weight: the muscle weight of MC group was (79.31±1.73) mg;TCM group was (107. 70±1. 57) mg;MPA group was (82.48+1. 52) mg. The muscle weight of TCM group was significantly higher than that of the MC and MPA groups(P=0. 005,P=0. 007). The tumor volumes: the tumor volumes of MC group was (7 664.34±286.00) mma ;TCM group was (3 935.44±561.40) mm3 ;MPA group was (5 960.22±290.00) mm:. The tumor volumes of TCM group was significantly lower than that of the MC and MPA groups(P--0. 005,P=0. 007). The tumor weight:the tumor weight of MC group was (8.06± 1.82) g;TCM group was (3.78±0.98) g;MPA group was (6.65+0.79) g. The tumor weight of TCM group was significantly lower than that of the MC and MPA groups(P = 0. 043, P=0. 052). There is no and difference between MC group ana MPA group. Life span:thelife spanof MC group was (29. 90±3.21) d;TCM group was (34. 80±3.39) d,MPA group was (30. 30± 3.20) d. The life span of TCM group was significantly longer than that of the MC and MPA groups (P=0. 043,P 0. 045). There was no any difference between MC group and MPA group(P=0. 307). CONCLUSlON:Bao Yuan detoxofi- cation soup is helpful to improve the life of quality and syndroma of lung cancer cachexia as well as to prolonge life span.
