肉毒梭菌培养液澄清过滤工艺的优化 被引量:1

Optimization of clarifying filtration procedure for Clostridium botulinum culture

摘要 目的优化肉毒梭菌培养液的澄清过滤工艺,并对过滤效果进行验证。方法选择不同型号的过滤器组合对A型肉毒梭菌培养液进行一级、二级澄清过滤,筛选最佳过滤器组合;用最佳过滤器组合对扩大培养批量的A、B、E、F型肉毒梭菌培养液进行澄清过滤。结果 P700K+50P过滤器组合对A型肉毒梭菌培养液进行一级、二级澄清过滤的效果最佳;用该过滤器组合对扩大培养批量的A、B、E、F型肉毒梭菌培养液进行澄清过滤,各型均可达到预期的试验目标。结论筛选出P700K+50P为A、B、E、F型肉毒梭菌培养液澄清过滤的最佳过滤器组合,为肉毒梭菌培养液澄清过滤工艺的进一步优化提供了实验依据。 Objective To optimize the clarifying filtration procedure for Clostridium botulinum culture. Methods Cl. bo- tulinum type A culture was subjected to clarifying filtrations of grades 1 and 2 by combination of various types of filters, based on which the optimal combination was screened. The Cl. botulinum types A, B, E and F in scale-up culture was subjected to clarifying filtration by the optimal combination. Results The combination of P700K + 50P used for clarifying filtrations of grades 1 and 2 of C1. butulinum type A showed satisfactory effect. Expected results were observed in the Cl. botulinum types A, B, E and F in scale-up culture after clarifying filtration by the optimal combination. Conclusion PT00K + 50P was screened as the optimal combination for clarifying filtration of Cl. botulinum types A, B, E and F, which provided an experimental basis for further optimization of clarifying filtration procedure for C1. botulinum culture.
