无机盐及葡萄糖对广州管圆线虫三期幼虫的体外趋化作用

In vitro chemotaxis of the third-stage larvae of Angiostrongylus cantonensis to inorganic salt and D-glucose

摘要 目的观察不同浓度的无机盐及葡萄糖对广州管圆线虫三期幼虫(AC-L3)的趋化作用。方法将含有20、50、100和150mmol/L无机盐(KCl、NaCl、Na2HPO4、NaHCO3、CaCl2、MgCl2)及葡萄糖溶液的纸片与蒸馏水浸泡的纸片分别贴于琼脂糖平皿的两个区域(实验区和对照区),将活力好的AC-L3离心后重悬,释放于平皿中央,避光孵化8h,显微镜下计数各区域幼虫。结果 20mmol/L Na2HPO4(P=0.016,CI=0.587)和50mmol/L NaCl(P=0.025,CI=0.696)对AC-L3有较明显的趋向作用,而100mmol/L KCl(P=0.027,CI=-0.546)和150mmol/L NaCl(P=0.028,CI=-0.366)对AC-L3有趋避作用。20mmol/L KCl、NaHCO3、MgCl2,100mmol/L CaCl2,150mmol/L NaHCO3、MgCl2均对AC-L3有趋向作用,但CI值均在0.5以下。不同浓度的葡萄糖对AC-L3的趋化作用差异无统计学意义(P>0.05)。结论 NaCl、NaHPO、KCl等无机盐对AC-L3具有一定的趋化作用,并且与无机盐的浓度相关。 Objective To examine the chemotatic response of the third-stage larvae of Angiostrongylus cantonensis (AC- L3) to inorganic salt and D-glucose. Methods Filter paper disks impregnated with inorganic salt (KC1,NaC1,Na2HPO4, NaHCO3,CaC12,MgC12) or D-glucose (20, 50, 100 and 150 mmol/L) were placed onto the agar plates containing 0.6% agarose. Active AC-L3s were placed on the centre of plates, and their migration was determined under a stereo microscope at 8 hours after hatching at dark. Results The larvae were found to migrate toward 20 mmol/L Na2HPO4 (P=0.016,CI= 0.587) and 50 mmol/L NaC1 (P=0.025,CI=0.696). On the contrary, the larvae were migrated away from the 100 mmol/L KCI (P=-0.027,CI=-0.546) and 150 mmol/L NaC1 (P=0.028,CI=-0.366). AC-L3s were also found to migrate toward 20 mmoL/L KC1, 20 mmol/L NaHCO3, 20 mmol/L MgC12, 100 mmol/L CaC12,150 mmol/L NaHCO3 and 150 mmol/L MgC12 (P〈0.05) ,respectively. But the CI-value was less than 0.5. D-glucose solution (20 mmol/L to 150 mmol/L) showed no activity to AC-L3s. Conclusion Third-stage larvae ofA. cantonensis exhibited chemotatic response to inorganic salts.
