摘要
目的:探讨脂肪干细胞(ADSCs)对糖尿病肾病大鼠氧化应激反应的抑制作用。方法:原代培养大鼠ADSCs,观察细胞形态,检测表面抗原。糖尿病肾病大鼠经尾静脉注射荧光标记的自体ADSCs,检测肾功指标,观察ADSCs在肾组织的分布及肾病理改变;测定肾组织氧化应激产物丙二醛及蛋白羰基含量,免疫印迹检测MnSOD和CuZnSOD的表达水平。结果:大鼠ADSCs类似成纤维细胞,呈旋涡状生长,高表达间充质干细胞标记CD90、CD44,内皮细胞标记CD31阴性,另外,CIN9d低表达,CD106阴性;ADSCs移植后糖尿病肾病大鼠的肾功生化指标明显恢复,肾组织内可见发红色荧光的ADSCs,病理形态明显改善,丙二醛、蛋白羰基含量及MnSOD、CuZnSOD表达量明显下调。结论:ADSCs可以通过抑制氧化应激反应治疗改善大鼠糖尿病肾病。
Objective: To investigate the inhibition of rat adipose derived stem cells (~s) on oxidative stress response of rats with diabetic nephropathy (DN). Methods: ~s of rats was primary cultured, cell morphology was observed, and surface antigens were identified by flow cytometry. Labeled ADSCs was transplanted into DN rats via tail vein. Renal biochemical parameters were detected. The distribution of ADSCs in the kidney tissues and pathological changes were observed; MDA and protein carbonyl content of oxidative stress in the kidneys was determined. MnSOD and CuZnSOD expression was detected by Western blotting. Results: Morphology of ADSCs was similar to fibroblasts with swirly growth, cDg0 and CD44 of mesenehymal stem cell markers was highly expressed. The endothelial cell marker CD31 was negative. While CIN9 was lowly expressed, and CD 106 was negative Renal biochemical indicators had been recovered after ADSCs was transplanted into DN rats. DSCs with red fluorescence were observed in the renal tissues. The renal pathological morphology was improved. The contents of MDA and protein carbonyl were reduced obviously, and the expression of MnSOD and CuZnSOD was significantly lower than that in DN rats. Conclusion: ADSCs can treat and improve rat with diabetic nephropathy by inhibiting oxidative stress.
