摘要
目的:研究实验性大鼠隐睾复位固定术对生精细胞凋亡及雌激素受体α(ERn)表达的影响。方法:建立大鼠左侧隐睾模型,模型建立7d后实施左侧隐睾复位固定术,采用TUNEL法检测睾丸生精细胞凋亡,免疫组织化学和RT-PCR检测各组睾丸ERα表达的变化。结果:隐睾内生精细胞凋亡增加,ERa表达降低;复位固定术能降低生精细胞凋亡,增强睾丸内ERα表达。复位固定术后24d生精细胞凋亡指数明显低于复位固定术前的隐睾组;复位固定术后48d生精上皮组织结构已基本恢复,其凋亡指数及ERα表达与假手术组比较,差异无统计学意义。结论:ERα与隐睾内生精细胞凋亡密切相关,隐睾复位固定术能通过恢复睾丸内ERα的表达,抑制生精细胞凋亡。
Objective: To investigate effect of orchidopexy on spermatogenic cell apoptosis and ERa expression in rat experimental cryptorchidism. Methods: Unilateral left cryptorchidism was surgically induced and the right side of testis was stayed into the scrotum after gubernaculum cut in each of 24 male SD rats (age of 21 days). On the 7th day after the establishment of cryptorchidism model, 8 rats were sacrificed (as Cryp group), and the others were operated with orchidopexy of left testis. On the 24th and 48th day after orchidopexy, 8 rats were sacrificed (as Oreh24 group and Orch48 group) respectively. The apoptosis of spermatogenic cell was detected by TUNEL and expression of ERa in testis was assessed using immunohistochemistry and RT-PCR assays. Results: Apoptosis of spermatogenic cell increased and expression of ERa reduced in cryptorchidism significantly. Orchidopexy suppressed apoptosis of spermatogenic cell and enhanced expression of ERα in testis. The apoptosis index in the Orch 24 group was less than that in the Cryp group, and there was no statistical difference in apoptosis index and expression of ERα between the Orch 48 group and Sham group. Conclusion: Orchidopexy can suppress sperrnatogenic cell apoptosis through regulating expression of ERa in the testis.
