摘要
目的:对水稻线粒体基因orf288功能进行初步探索。方法:笔者在大肠杆菌中构建水稻线粒体基因orf288的原核表达载体pET-30a-orf288,转化入BL21(DE3)后利用IPTG诱导,绘制生长曲线,分析其表达产物对细菌生命活动的影响。结果:成功构建了pET-30a-orf288重组子,并能成功表达ORF288。结论:原核表达产物ORF288对细菌的生长具有明显的抑制作用,为进一步研究orf288作用提供了研究线索。
Objective: The function of rice mitochondrial gene orf288 in a preliminary exploration. Method: To study the molecular mecha-nism of this gene,the sequence of orf288 gene was cloned into pET- 30a, and then expressed in BL21 (DE3) strain. Result: The results showed that weakly expressed native ORF288 proteins induced from the recombinant pET - 30a - orf288 strongly inhibit the growth of E-.coli. Conclusion: These results may provide a clue to get some information about the mechanism of orf288.
